Knock-in (Knock-in, KI) is a technique that utilizes homologous recombination of genes to transfer an exogenous functional gene into a homologous sequence in the cell and genome, and obtain expression well in the cell after gene recombination.

It takes 5-6 months to obtain Neo-deleted heterozygous mice and a total of 10-12 months to construct a KI homozygous mouse models with low success rates using traditional techniques, because these techniques have low chimera positivity and require mating with a mouse to obtain Neo-deleted heterozygous mice, and then breeding the born heterozygous mice for 2-3 generations to get the homozygous KI mice.
